I like twhirl. I used tweetdeck for a while and it was very buggy.

I think that the best interface goes to Tweetie, but I cannot use it in Tiger it seems which sucks. I am using Tweetr, can't stand the other apps interfaces.
I really enjoy Tweetie for the Mac. It does anything your typical Twitter client would do, but it has a great interface and it's very easy to pick up on.

I really like using Twhirl for the mac. very simple and easy to use interface, despite having plenty of features and settings so you can fine tune it to want you want it to do.
Best one I've used on Windows Vista is definitely Twhirl. I never liked the interface of TweetDeck, and Twhirl is just right for me. Plus, the themes are awesome.

Tweetie has an AWESOME UI, and I'm using it currently, but the only issue I have with it is like a lot of the time when I try to post something, it says- Cannot Post Tweet and then (null) under it. =/ Tweetie need to fix some of the minor bugs they have in the next version!
